چکیده

Background: Among anti-hypertension drugs, calcium (Ca) antagonists cause gingival overgrowth as a side effect. We previously discovered that this side effect was due to elevation of the calcium concentration in the cytosol ([Ca]i). Ca entry through non-selective cation channels (NSCCs) and Ca release from intracellular Ca stores are involved in this [Ca]i elevation. Furthermore, we discovered that calcium-sensing receptors (CaSRs) participate in nifedipineinduced [Ca]i elevation. Transient receptor potential (TRP) channels have been identified as NSCCs. In the present study, we undertook experiments to determine if TRPV1 channels are present in gingival fibroblasts and to ascertain if nifedipine-activated NSCCs are TRPV1 channels. Methods: Normal human gingival fibroblast Gin-1 cells were used. The [Ca]i was measured using a video-imaging analysis system with the Ca-sensitive fluorescent dye fura-2/AM. Results: The NSCC inhibitor SKF96365 significantly inhibited nifedipine-induced [Ca]i elevation. TRPV1 channel agonists such as capsaicin, olvanil and resiniferatoxin concentration-dependently elevated the [Ca]i. The TRPV1 channel activator anandamide concentration-dependently increased the [Ca]i. The TRPV1 channel antagonists capsazepine, AMG9810, iodoresiniferatoxin, ruthenium red, and SB366791 significantly inhibited nifedipine-induced [Ca]i elevation. Conclusion: These results suggest that Ca entry through TRPV1 channels is involved in the nifedipine-induced [Ca]i elevation seen in gingival fibroblasts. We describe here a modified version of our “calcium trigger theory”.

Multiple H+ sensors mediate the extracellular acidification-induced [Ca2+]i elevation in cultured rat ventricular cardiomyocytes

Acidosis has been known to cause "Ca2+ transients", however, the mechanism is still uncertain. Here, we demonstrated that multiple H+ sensors, such as ASICs, TRPV1 and proton-sensing G protein coupled receptors (GPCRs) are involved in extracellular acidification-induced intracellular calcium ([Ca2+]i) elevation. By using calcium imaging measures, we observed that both ASIC and TRPV1 channels in...

Gestational hypothyroidism-induced changes in L-type calcium channels of rat aorta smooth muscle and their impact on the responses to vasoconstrictors

Objective(s): Thyroid hormones play an essential role in fetal growth and maternal hypo-thyroidism which leads to cardiovascular deficiency in their offspring.  Considering this, we intended to investigate the impact of gestational hypothyroidism on offspring vascular contractibility and possible underlying mechanisms. Materials and Methods: Hypothyroidism was induced in female rats by administ...

Characterization of voltage-sensitive calcium channels in growth plate chondrocytes.

Growth plate chondrocytes (GPCs), cells integrally involved with the process of endochondral bone formation, facilitate Ca2+ infux to provide a source of ion for processes such as Ca2+ signaling and matrix vesicle loading. We hypothesize that this Ca2+ entry into GPCs is achieved through the action of voltage-sensitive Ca2+ channels. This hypothesis was tested by measuring intracellular [Ca2+] ...

مقایسه اثر وراپامیل، نیفدیپین و دیلتیازم بر آستانه تشنجات کلونیک ناشی از پنتیلن تترازول در موش سوری

Background & Objective: Verapamil, nifedipine and diltiazem are calcium channel blockers widely used as a variety of cardiovascular ailment in humans. A number of studies have shown that calcium channel blockers have anticonvulsant effect in a range of animal seizure models (but not all animals). The aim of this study was to investigate the effects of verapamil, nifedipine and diltiazem on pe...

Privileged crosstalk between TRPV1 channels and mitochondrial calcium shuttling machinery controls nociception.

The nociceptive noxious heat-activated receptor - TRPV1, conducts calcium and sodium, thus producing a depolarizing receptor potential, leading to activation of nociceptive neurons. TRPV1-mediated calcium and sodium influx is negatively modulated by calcium, via calcium-dependent desensitization of TRPV1 channels. A mitochondrial Ca2+ uniporter - MCU, controls mitochondrial Ca2+ entry while a s...
